目的 研究中华芦荟多糖 (AP)对X射线照射后人非瘤细胞株CLiver和 2 93细胞凋亡及细胞凋亡蛋白 p5 3,bax ,Bcl 2 ,Bad ,survivin表达的影响。方法 流式细胞仪检测细胞凋亡 ,Westernblot检测细胞凋亡相关蛋白的表达。结果 X射线照射后 72h ,CLiver细胞可见典型的DNA梯形条带 ,照射前加入AP 2 5或 5 0mg·L-1均能使DNA片段减少 ;AP预处理使CLiver细胞在照后 6h和72h凋亡率分别由 5 0 %和 4 3 0 %降至 2 2 %和 10 9% ,使2 93细胞照后 6h凋亡率由 8 6 %降至 4 0 % ;AP预处理可使CLiver细胞p5 3、Bad、Bax蛋白表达量下调 ,Bcl 2和sur vivin蛋白表达则上调。结论 AP对X射线辐射后非瘤细胞凋亡的抑制作用与对凋亡蛋白表达的调节有关
Objective To study the effect of aloe polysaccharides (AP) on the apoptosis and the expression of apoptosis proteins p53, bax, Bcl2, Bad and survivin in human non-neoplastic cell lines (Chriver and 293) after X-irradiation. Methods Apoptosis was detected by flow cytometry. The expression of apoptosis-related proteins was detected by Western blot. RESULTS: After 72h of X-irradiation, typical DNA trapezoid bands were seen in the CLiver cells. Addition of AP 25 or 50 mg·L-1 before irradiation could reduce DNA fragmentation; AP pretreatment caused the CLiver cells to wither at 6h and 72h after irradiation. Mortality rates dropped from 50% and 40% to 22.2% and 109%, respectively, and the rate of apoptosis decreased from 86% to 40% at 293h after irradiation in 293 cells. AP pretreatment could make CLiver cells p5 3, Bad, Bax protein expression was down-regulated, Bcl 2 and sur vivin protein expression was upregulated. Conclusion The inhibitory effect of AP on the apoptosis of non-neoplastic cells after X-irradiation is related to the regulation of apoptosis protein expression
