目的研究胃转流术对2型糖尿病大鼠肝脏FoxO1表达的影响。方法 40只SD大鼠随机分为糖尿病手术组(A组)、假手术组(B组)、正常对照组(C组)、糖尿病对照组(D组),每组10只。C、D组按A、B组术前和术后4、8、16、24周检测空腹血糖、空腹和餐后血清胰岛素、胰高血糖素样肽(GLP-1)。24周后处死各组大鼠,免疫组化和Western blot检测各组肝脏中FoxO1的表达。结果 A组术后4周空腹血糖由术前的(20.2±2.1)mmol/L降至(10.5±2.5)mmol/L,术后24周空腹血糖为(7.7±1.1)mmol/L,术后4周空腹胰岛素由术前(2.87±0.08)mU/L增至(2.91±0.08)mU/L,术后24周空腹胰岛素为(3.00±0.06)mU/L;术后4周空腹血清GLP-1由术前(8.7±0.9)pmol/L升至(13.2±1.0)pmol/L,术后24周为(23.5±0.2)pmol/L。A组术后24周肝脏FoxO1表达量为18.4±1.2,B组为37.6±0.5,C、D两组分别为9.2±1.0、35.8±0.4,A组术后FoxO1的表达量明显低于B、D组,差异有统计学意义(P<0.05)。A组血清GLP-1浓度与肝脏FoxO1的表达量成负相关(r=-0.864,P<0.05)。结论胃转流术可显著降低2型糖尿病大鼠肝脏FoxO1的表达,降低血糖。
Objective To investigate the effect of gastric bypass on the expression of FoxO1 in the liver of type 2 diabetic rats. Methods Forty SD rats were randomly divided into diabetic surgery group (A group), sham operation group (B group), normal control group (C group) and diabetic control group (D group). Groups C and D were given fasting blood glucose, fasting and postprandial serum insulin and glucagon-like peptide (GLP-1) in groups A and B at 4, 8, 16 and 24 weeks after operation. After 24 weeks, the rats in each group were sacrificed, and the expression of FoxO1 in each group was detected by immunohistochemistry and Western blot. Results The fasting blood glucose in group A decreased from (20.2 ± 2.1) mmol / L preoperatively to (10.5 ± 2.5) mmol / L 4 weeks after operation, and (7.7 ± 1.1) mmol / L at 24 weeks postoperatively The fasting insulin concentration at 4 weeks increased from (2.87 ± 0.08) mU / L to (2.91 ± 0.08) mU / L and fasting insulin at 24 weeks postoperatively was (3.00 ± 0.06) mU / L. The fasting serum GLP- 1 increased to (13.2 ± 1.0) pmol / L preoperatively from (8.7 ± 0.9) pmol / L and (23.5 ± 0.2) pmol / L at 24 weeks postoperatively. The expression of FoxO1 in group A at 24 weeks after operation was 18.4 ± 1.2, in group B was 37.6 ± 0.5, in group C and D was 9.2 ± 1.0 and 35.8 ± 0.4 respectively. The expression of FoxO1 in group A was significantly lower than that in group B D group, the difference was statistically significant (P <0.05). A group of serum GLP-1 concentration and liver FoxO1 expression was negatively correlated (r = -0.864, P <0.05). Conclusion Gastric bypass can significantly reduce the expression of FoxO1 in the liver of type 2 diabetic rats and lower the blood sugar.
