慢性乙型肝炎病毒(hepatitis B virus,HBV)感染影响着全球近2.5亿人的健康。HBV共价闭合环状DNA(HBV cccDNA)在病毒复制中发挥关键作用,由于HBV cccDNA以微染色体的形式存在于被感染的肝细胞核内,无法被有效清除,导致了病毒感染的持续,这也是目前慢性乙型肝炎治疗的难点。表观遗传是基因表达调控领域的重大发现,其中组蛋白修饰(乙酰化、甲基化、泛素化等)所形成的组蛋白密码更是展现了表观遗传调控基因表达的精细复杂。本文拟就组蛋白修饰对HBV cccDNA的调控作用作一综述。
Chronic hepatitis B virus (HBV) infection affects the health of nearly 250 million people worldwide. HBV covalently closed circular DNA (HBV cccDNA) play a key role in viral replication. Since cccDNA is present in the infected hepatocyte nucleus in the form of minichromosomes, it can not be effectively eliminated, which leads to the persistence of virus infection The current treatment of chronic hepatitis B difficult. Epigenetics is a major discovery in the field of regulation of gene expression, in which the histone code formed by histone modifications (acetylation, methylation, ubiquitination, etc.) further demonstrates the subtle sophistication of epigenetic regulation of gene expression. In this paper, the regulatory effect of histone modifications on HBV cccDNA is reviewed.
