目的:探讨奥曲肽抑制前列腺癌生长的药理作用与多药耐药现象之间的相关性。方法:MTT比色分析法测定奥曲肽对人前列腺癌细胞株PC-3生长的影响;实时定量PCR法,分别检测PC-3细胞P糖蛋白(P-gp)和多药耐药相关蛋白(MRP)的mRNA水平。结果:1×10~(-9)~1×10~(-6)mol·L~(-1)的奥曲肽能呈浓度依赖性抑制PC-3细胞的增殖,同时下调MRP mRNA的表达水平。但PC-3细胞检测不到P-gP mRNA的表达。结论:奥曲肽抑制PC-3细胞的增殖可能与其抑制MRP-3的表达相关。
Objective: To explore the correlation between the pharmacological effects of octreotide on inhibiting the growth of prostate cancer and multidrug resistance. Methods: MTT assay was used to determine the effect of octreotide on the growth of human prostate cancer cell line PC-3. Real-time quantitative PCR was used to detect the expression of P-gp and multidrug resistance-related protein (MRP) ) MRNA levels. Results: Octreotide inhibited the proliferation of PC-3 cells in a concentration-dependent manner and down-regulated the expression of MRP mRNA in the concentration range of 1 × 10 -9 ~ 1 × 10 -6 mol·L -1. However, P-gP mRNA expression was undetectable in PC-3 cells. Conclusion: Octreotide inhibits the proliferation of PC-3 cells may be related to the inhibition of MRP-3 expression.
