目的:探讨组织蛋白酶L(Cathepsin L,CTSL)与组织蛋白酶B(Cathepsin B,CTSB)在子痫前期胎盘滋养细胞异常表达的情况。方法:选取40例子痫前期、40例早发型子痫前期孕妇为研究对象,另选取40例正常健康孕妇为对照组,采用免疫组化法测定各组孕妇血清CTSL、CTSB表达情况及采用RT﹣PCR法测定各组CTSL、CTSB的表达量。结果:与对照组相比,子痫前期、早发型子痫患者CTSL、CTSB阴性表达率较高,其中早发型子痫患者CTSL、CTSB阴性表达率高于子痫前期,两组比较差异有统计学意义(P<0.05)。对照组CTSLmRNA、CTSBmRNA表达量显著高于子痫前期及早发型子痫前期患者,而早发型子痫患者则低于子痫前期,差异均有统计学意义(P<0.05)。结论:人类滋养层细胞CTSL、CTSB异常表达与子痫的发生关系密切,通过测定子痫患者CTSL、CTSB水平可有效预测患者病情进展。
Objective: To investigate the abnormal expression of placental trophoblasts in preeclampsia of Cathepsin L (CTSL) and Cathepsin B (CTSB). Methods Forty pregnant women with preeclampsia and 40 early pregnant women with preeclampsia were enrolled in this study. Forty healthy healthy pregnant women were selected as control group. The expression of CTSL and CTSB in serum of pregnant women in each group were determined by immunohistochemistry and RT- PCR method to determine the expression of CTSL, CTSB. Results: Compared with the control group, the positive rates of CTSL and CTSB in preeclampsia were higher than those in the control group, and the positive rate of CTSL and CTSB in preeclampsia was higher than that in preeclampsia Significance (P <0.05). The levels of CTSLmRNA and CTSBmRNA in control group were significantly higher than those in preeclampsia and early-onset preeclampsia, while those in early onset eclampsia were lower than those in preeclampsia (P <0.05). Conclusion: Abnormal expression of CTSL and CTSB in human trophoblastic cells is closely related to the occurrence of eclampsia. The determination of CTSL and CTSB in patients with eclampsia can effectively predict the progression of the disease.
