目的探讨难治性癫痫患者颞叶皮质中二肽基肽酶6(DPP6)的表达情况。方法在重庆医科大学建立的脑组织库中,按照随机化原则抽取27例癫痫患者的脑组织(癫痫组)和15例脑外伤患者的脑组织(对照组),应用免疫组织化学染色、蛋白质印迹杂交、RT-PCR法检测DPP6的表达水平。结果 DPP6表达于两组大脑神经元的细胞膜,蛋白质印迹杂交结果显示,癫痫组和对照组DPP6的平均光密度(OD)值分别为0.86±0.04和0.37±0.02,差异有统计学意义(P<0.001)。癫痫组和对照组脑组织DPP6 mRNA的相对表达量分别为0.74±0.03和0.35±0.03,差异有统计学意义(P<0.001)。结论异常表达的DPP6可能参与了难治性癫痫的形成。
Objective To investigate the expression of dipeptidyl peptidase 6 (DPP6) in temporal cortex of refractory epilepsy patients. Methods Brain tissue (epilepsy group) and brain injury group (control group) were collected from 27 patients with epilepsy in accordance with the principle of randomization in the brain tissue bank established by Chongqing Medical University. Immunohistochemical staining, Western blotting Hybridization, RT-PCR method to detect DPP6 expression levels. Results DPP6 was expressed on the cell membrane of neurons in both groups. The results of Western blotting showed that the average optical density (OD) values of DPP6 in epilepsy group and control group were 0.86 ± 0.04 and 0.37 ± 0.02, respectively (P < 0.001). The relative expression levels of DPP6 mRNA in epilepsy group and control group were 0.74 ± 0.03 and 0.35 ± 0.03, respectively, with statistical significance (P <0.001). Conclusion Abnormal expression of DPP6 may be involved in the formation of refractory epilepsy.
