介绍了3种同孔径不同键合基团的大孔径硅胶聚合物键合相(HDSI,PAMSI,PFMSI)的合成,3种填料是在 100 nm孔径硅胶上键合乙烯基后,分别与甲基丙烯酸羟乙基酯、二乙烯基苯共聚形成HDSI填料;与甲基丙烯酸胺共聚形成PAMSI填料;与顺丁烯二酸共聚再与乙二胺反应形成PFMSI填料。比较了它们对蛋白质分离的特性。结果表明:3种填料都能达到蛋白质快速分离。并且讨论了在PAMSI色谱柱上,盐浓度对蛋白质分离的影响;在PFMSI色谱柱上,流动相pH对蛋白质分离的影响
The synthesis of three kinds of macroporous silica gel polymer bonded phases (HDSI, PAMSI, PFMSI) with different bonding groups with the same pore diameter was introduced. The three kinds of fillers were synthesized by bonding vinyl groups on 100 nm silica gel, Hydroxyethyl acrylate and divinylbenzene to form HDSI filler; copolymerizing with methacrylamide to form PAMSI filler; copolymerizing with maleic acid and then reacting with ethylenediamine to form PFMSI filler. Compared their characteristics of protein separation. The results showed that all three kinds of fillers can achieve rapid protein separation. The influence of salt concentration on protein separation on PAMSI was also discussed. The influence of mobile phase pH on protein separation on PFMSI column
