我们曾证实局部麻醉药布比卡因(bupivacaine)与加温合并应用后可增加肿瘤细胞的热杀伤效应。为进一步探讨不同序贯处理的热增敏差异并为今后临床治疗方案的设计提供理论依据,本实验观察了不同给药顺序对细胞热敏感性的影响,初步结果证实先给药后加温序贯处理可达最大热增敏效应。 实验所用细胞为人宫颈癌细胞系Hela,细胞浓度为1×10~6/ml。将等量接种的细胞分为5组:(1)正常对照组(C组);(2)药物组(B组),每毫升细胞悬液中加入50μg布比卡因,37℃作用60分钟;(3)加温组(H组),44℃,30-分钟水浴加温;(4)先给药后加温组(BH组);(5)先加温后给药组(HB组)。观察指标为细胞生长曲线(连
We have demonstrated that local anesthetic bupivacaine combined with warming can increase the heat killing effect of tumor cells. In order to further explore the differences in heat sensitization of different sequential treatments and provide theoretical basis for the design of clinical treatment plans in the future, this experiment observed the effects of different dosing orders on cell thermal sensitivity. The preliminary results confirmed the sequential administration of warming after the first administration. The maximum thermal sensitizing effect can be achieved. The cells used in the experiment were human cervical cancer cell line HeLa, and the cell concentration was 1×10 6 /ml. Equally vaccinated cells were divided into 5 groups: (1) normal control group (C group); (2) drug group (B group), 50 μg of bupivacaine per ml of cell suspension was added and incubated at 37°C for 60 minutes.
