目的评价CpG佐剂对以PfCP-2.9/ISA720佐剂为基础的联合疟疾抗原的免疫原性。方法使用ISA720及ISA720+CpG佐剂与恶性疟原虫抗原制备制剂,免疫BALB/c小鼠,考察不同佐剂增强免疫的效果。通过检测免疫血清中特异性IgG的水平、识别天然抗原的能力、分析特异性IgG的亚类分布等指标进行评价。结果 CpG佐剂显著提升了联合疟疾抗原在BALB/c小鼠中产生特异性抗体的量(P<0.01),免疫血清能有效识别红内期疟原虫天然抗原,IFA滴度显著提高(P<0.01),IgG亚类分析显示该佐剂能有效刺激小鼠产生针对PfCP-2.9和Pfclet-3的多种IgG亚类抗体。结论 CpG显著提高了联合疟疾抗原的免疫原性。
Objective To evaluate the immunogenicity of CpG adjuvants against the combined malaria antigen based on the PfCP-2.9 / ISA720 adjuvant. Methods BALB / c mice were immunized with ISA720 and ISA720 + CpG adjuvant and Plasmodium falciparum antigens to study the effect of different adjuvants to enhance immunity. By detecting the level of specific IgG in immune serum, the ability to recognize natural antigens, and the analysis of subgroup distribution of specific IgG, we evaluated them. Results CpG adjuvant significantly increased the amount of specific antibodies produced by BALB / c mice in combination with malaria antigen (P <0.01). The immune serum could effectively identify the natural antigens of Plasmodium falciparum and IFA titer was significantly increased (P < 0.01). IgG subclass analysis showed that the adjuvant effectively stimulated mice to produce various IgG subclass antibodies against PfCP-2.9 and Pfclet-3. Conclusion CpG significantly increased the immunogenicity of the combined malaria antigen.
