目的:观察加味四逆散对肝脏Ⅳ型胶原(C0L-Ⅳ)和转化生长因子β1(TGFβ1)mRNA的影响,探讨其抗肝纤维化的作用机制。方法:将50只动物购回后随机分为4组,除空白对照组10只外,其余40只均用二甲基亚硝胺(DMN,用生理盐水稀释至5 g.L-1)以10 mL.kg-1体重ip,每日1次,每周连续3 d,连续4周的方法制备大鼠肝纤维化模型,加味四逆散按生药量以35 g.kg-1ig给药,采用实时定量PCR技术检测各组大鼠肝组织Ⅳ型胶原和TGFβ1 mRNA表达的水平。结果:加味四逆散组大鼠肝组织C0L-ⅣmRNA为5.136±1.32,TGFβ1 mRNA为4.839±0.48,与病理模型组(C0L-ⅣmRNA 10.369±1.69,TGFβ1mRNA 6.307±0.85)比较有显著性差异(P<0.05)。结论:加味四逆散能够抑制肝纤维化大鼠肝组织C0L-Ⅳ和TGFβ1 mRNA表达,从而发挥其抗肝纤维化的作用。
Objective: To observe the effect of Jiawei Sini Decoction on the expression of collagen type Ⅳ (C0L-Ⅳ) and transforming growth factor-β1 (TGFβ1) in liver and its mechanism of anti-hepatic fibrosis. Methods: Fifty animals were purchased and randomly divided into four groups. Except for the blank control group of 10, the remaining 40 animals were treated with dimethylnitrosamine (DMN, diluted to 5 gL-1 with normal saline) and 10 mL .kg-1 weight ip, once a day, 3 days a week for 4 weeks to prepare a rat model of liver fibrosis, plus Sini powder according to the amount of raw drugs to 35 g.kg-1ig administration, using real-time Quantitative PCR was used to detect the expression of type Ⅳ collagen and TGFβ1 mRNA in the liver of each group. Results: The C0L-ⅣmRNA and the mRNA expression of TGFβ1 in Jiawei Sini San group were 5.136 ± 1.32 and 4.839 ± 0.48, respectively, which were significantly different from those in pathological model group (C0L-ⅣmRNA 10.369 ± 1.69 and TGFβ1mRNA6307 ± 0.85) (P <0.05). Conclusion: Jiawei Sini Decoction can inhibit the expression of C0L-Ⅳ and TGFβ1 mRNA in liver tissue of rats with hepatic fibrosis, and thus exert its anti-hepatic fibrosis effect.
