Spred家族是一组Sprouty相关的酪氨酸激酶结合蛋白,它对多种生长因子诱导的Ras-ERK信号途径有负调控作用。Spred家族各成员在染色体上定位和体内表达均不相同,但都具有保守的结构域:C端Sprouty相关结合域(SPR)、中央c-Kit结合域(KBD)和N端Enabled/VASP同源结构域(EVH-1)。Spred调节细胞发育、运动以及增殖等生命活动,参与肿瘤的转移、造血调控、过敏反应以及骨的形成等病理生理学过程。本文主要综述Spred家族的结构及功能特点以及对细胞增殖信号途径的调节作用。
The Spred family is a group of Sprouty-associated tyrosine kinase binding proteins that negatively regulate the various growth factor-induced Ras-ERK signaling pathways. Each member of the Spred family has different chromosomal location and in vivo expression but all have conserved domains: the C-terminal Sprouty-associated domain (SPR), the central c-Kit binding domain (KBD) and the N-terminal Enabled / VASP homolog Domain (EVH-1). Spred regulates the life activities such as cell development, movement and proliferation, and participates in pathophysiological processes such as tumor metastasis, hematopoietic regulation, allergic reaction and bone formation. This review summarizes the structural and functional characteristics of the Spred family and its regulatory role in cell proliferation signaling pathways.
