本文报告了应用 ̄32P和光敏生物素标记克隆的pBF4DNA,作为探针,以斑点杂交试验,检测海南、云南和山东省的血样。两种标记探检测结果与镜检的符合率分别为恶性疟98.6%和96.2%;间日疟91.6%和86.7%;正常人血95.2%和100%;并可从多种疟原虫标本中,特异地检出恶性疟原虫,显示探针具有恶生疟原虫的种特异性。检测的敏感度为84个原虫/μl血,检测原虫DNA可达10pg水平。表明探针具有良好的实用性。
In this paper, pBF4 DNA labeled with biotinylated biotin was reported as a probe to detect blood samples from Hainan, Yunnan and Shandong provinces by dot blot hybridization. The coincidence rates of the two kinds of marker detection and microscopic examination were 98.6% and 96.2% of P. falciparum respectively; 91.6% and 86.7% of P. vivax; 95.2% and 100% of normal human blood; The Plasmodium falciparum can also be detected specifically from a wide range of Plasmodium specimens, demonstrating that the probe has the species specificity of Plasmodium falciparum.
