目的 建立和优化蛋白质分子差异展示的双向聚丙烯酰胺凝胶电泳技术 ,初步观察巴豆提取物诱导小鼠小肠组织中蛋白质的差异表达 ,为进一步筛选其中介导巴豆生物作用的蛋白质分子奠定基础。方法 应用巴豆提取物灌胃制备小鼠慢性胃肠运动增强模型 ,提取其小肠组织中总蛋白质 ,优化蛋白质分子差异展示的双向聚丙烯酰胺凝胶电泳技术 ,并用其分析模型动物小肠组织中蛋白质 ,双向电泳凝胶银染法显示其蛋白质的差异表达。结果 建立了小鼠慢性胃肠运动增强模型 ,优化并运用双向聚丙烯酰胺凝胶电泳技术展示出模型动物小肠组织中有差异蛋白质的表达。结论 巴豆提取物可以诱导小鼠小肠组织中蛋白质差异表达 ,这些差异表达的蛋白质可能介导了巴豆的生物作用
Objective To establish and optimize the two-dimensional polyacrylamide gel electrophoresis technique for differential display of protein molecules, and initially observe the differential expression of proteins in small intestine of mice induced by croton extract, so as to lay the foundation for further screening of protein molecules that mediate the biological action of Croton. Methods The gastrointestinal motility enhancement model was established by intragastric administration of croton extract, and the total protein in the small intestine was extracted. The two-dimensional polyacrylamide gel electrophoresis technique for optimizing the protein molecular display was used to analyze the proteins in the small intestine of model animals. Two-dimensional electrophoresis gel silver staining showed differential expression of the protein. Results A mouse model of chronic gastrointestinal motility enhancement was established. Two-dimensional polyacrylamide gel electrophoresis was used to optimize the expression of differential proteins in the small intestine of model animals. Conclusion The croton extract can induce differential expression of proteins in the small intestine of mice. These differentially expressed proteins may mediate the biological effects of Croton.
