目的:观察藤茶总黄酮(Tengcha flavonoids,TCF)对大鼠实验性肝纤维化的治疗作用,并探讨其机制。方法:采用四氯化碳(CCL4)皮下注射制备大鼠肝纤维化模型,随机分为正常对照组、模型对照组、藤茶总黄酮(TCF)治疗组和秋水仙碱(CLC)治疗组。各组以灌胃的形式给药,采用蛋白免疫印迹分析和实时定量PCR技术检测各组大鼠肝组织Ⅰ、Ⅲ型胶原表达情况。结果:藤茶总黄酮能明显降低实验性大鼠肝组织中Ⅰ、Ⅲ型胶原的含量,与模型对照组比较有显著性差异(P<0.05)。结论:藤茶总黄酮可通过减少肝组织Ⅰ、Ⅲ型胶原的生成,发挥抗肝纤维化的作用。
Objective: To observe the therapeutic effect of Tengcha flavonoids (TCF) on experimental liver fibrosis in rats and to explore its mechanism. METHODS: Hepatic fibrosis model was established by subcutaneous injection of carbon tetrachloride (CCL4) and randomly divided into normal control group, model control group, total flavonoids (TCF) treatment group and colchicine (CLC) treatment group. Each group was administered intragastrically and Western blot analysis and real-time quantitative PCR were used to detect the expression of type I and type III collagen in rat livers. RESULTS: Total flavonoids of vine tea could significantly reduce the content of type I and type III collagen in experimental rat liver tissue, and there was a significant difference compared with the model control group (P<0.05). Conclusion: Total flavonoids from vine tea can reduce the formation of type I and type III collagen in liver tissue and exert its anti-fibrotic effect.
