切取双色花叶芋(Caladium bicolor)愈伤组织为外植体,分别置于相同辐照度(≈3.2w/m~2)的白(W)、红(R)、黄(Y)、绿(G)、蓝(B)光及黑暗(D)中,比较不同光质对双色花叶芋组织培养形态发生的影响。结果表明:1.不同光质处理并未引起苗的形态发生途径产生差异(发生不定芽),但能影响培养物的生物总量、器官发生的先后和多寡。其中黄光诱导芽发生的频率最高。2.测定叶绿素a、叶绿素b及类胡萝卜素的含量,发现黄光处理的样品,其含量最高;红光、绿光及蓝光稍逊于白光;黑暗处理对色素的合成和积累最为不利。3.通过对再生植株叶片的电镜观察,发现不同光质对于叶片中叶绿体的亚显微结构影响显著。
Cut callus of Caladium bicolor as explants were placed in the same irradiance (≈3.2w / m ~ 2) of the white (W), red (R), yellow (Y), green (G), blue (B) light and dark (D), and compared the effects of different light qualities on the morphology of the tissue culture of the two-colored Cucurbita. The results showed that: 1. Different light quality treatments did not cause morphological differences in seedling emergence (adventitious buds), but could affect the total biomass of the culture, the organ occurrence and the amount. Among them, Huangguang induced the highest frequency of buds. The contents of chlorophyll a, chlorophyll b and carotenoid were determined. The results showed that the content of chlorophyll a, chlorophyll b and carotenoid was the highest in the samples treated with yellow light, the light in red, green and blue was less than that in white. Through the electron microscope observation of regenerated plant leaves, it was found that different light quality had a significant effect on the sub-microstructure of chloroplasts in leaves.
