目的观察四藤方中中药及联合应用对人胃癌SGC-7901细胞增殖、凋亡的影响。方法体外培养胃癌SGC-7901细胞,不同浓度中药作用后,CCK-8法检测细胞增殖,流式细胞仪检测细胞凋亡;Caspase-3活性检测试剂盒检测SGC-7901细胞Caspase-3的活性。结果与对照组比较,四藤方中菝葜、红藤、藤梨根均能抑制SGC-7901细胞增殖。菝葜、红藤、藤梨根联用可增强对SGC-7901细胞增殖和凋亡的作用,同时提高SGC-7901细胞Caspase-3活性(P<0.01)。结论菝葜、红藤、藤梨根三药联合使用可增强对人胃癌SGC-7901的抗癌作用,并可能与活化Caspase-3介导细胞凋亡相关。
Objective To observe the effect of Si Teng Fang Zhong Chinese medicine and its combination on proliferation and apoptosis of human gastric cancer cell line SGC-7901. Methods SGC-7901 cells were cultured in vitro. Cell proliferation was detected by CCK-8 assay and apoptosis was detected by flow cytometry. The activity of Caspase-3 in SGC-7901 cells was detected by Caspase-3 activity assay kit. Results Compared with the control group, Si Teng Fang midge, red vine, rattan pear root can inhibit the proliferation of SGC-7901 cells. Scirpus mandshurica, Radix et Rhizoma corydalis and ramie could increase the proliferation and apoptosis of SGC-7901 cells and increase the activity of Caspase-3 in SGC-7901 cells (P <0.01). CONCLUSION: The combined use of three Chinese medicinal plants, Scrophulariaceae, Tengli and Tengligen can enhance the antitumor effect of SGC-7901 on human gastric cancer cells and may be related to the activation of Caspase-3-mediated apoptosis.
