目的探讨黄芩苷对体外培养的人毛囊生长、毛乳头细胞增殖和分泌血管内皮生长因子(VEGF)的影响。方法选择不同浓度黄芩苷作用于体外培养的人头皮毛囊8天,观察人毛囊生长及形态变化:作用于人毛乳头细胞72 h,四甲基偶氮唑监(MTF)法测定细胞增殖活性,ELISA法检测细胞分泌的VEGF水平。结果7.5.15,30,45μg/mL的黄芩苷对体外培养的人头皮毛囊有明显促生长作用;3.75,7.5,15,30μg/mL的黄芩苷对毛乳头细胞增殖无明显影响,但促进细胞分泌VEGF,与阴性对照组相比,差异有统计学意义。结论黄芩苷促进体外培养的人头皮毛囊生长,部分可能是通过增加毛乳头细胞分泌VEGF促使毛发生长。
Objective To investigate the effects of baicalin on growth of human hair follicles, proliferation of dermal papilla cells and secretion of vascular endothelial growth factor (VEGF) in vitro. Methods Human scalp hair follicles were cultured in vitro with baicalin at different concentrations for 8 days. Growth and morphological changes of human hair follicles were observed. Human hair papilla cells were treated for 72 h, and cell proliferation activity was measured by MTF assay. The level of VEGF secreted by cells was measured by ELISA. RESULTS Baicalin at 7.5.15, 30, 45 μg/mL had a significant growth-promoting effect on human scalp hair follicles in vitro; baicalin at 3.75, 7.5, 15 and 30 μg/mL had no obvious effect on the proliferation of dermal papilla cells, but promoted the cells. Secretion of VEGF was statistically significant compared with the negative control group. Conclusion Baicalin can promote the growth of human scalp hair follicles in vitro, and may partly promote hair growth by increasing VEGF secretion from dermal papilla cells.
