目的研究广西永福县长寿老人载脂蛋白E(ApoE)基因多态性与永福长寿现象的关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术,对319名年龄90~113岁的广西永福县长寿老人和168名作为相应年龄对照的长寿家庭成员,并以当地非长寿自然人群104人作为对照组,进行ApoE基因分型,并分别比较长寿老人与各组人群的基因型、基因频率的分布特征。结果 (1)广西永福县长寿老人ApoE基因型频率中以ε3/3最高,其次是ε2/3和ε3/4,而ε2/4和ε4/4频率最少。长寿老人组与其亲属年龄对照的ApoE基因型和基因频率相比之间均无显著性差别。(2)长寿组与当地自然人群对照相比ε3/ε3(82.4%;75.0%)基因型和ε3等位基因(90.44%;85.17%)呈显著性阳性关联(P<0.05,P<0.01)。结论 ApoE基因多态性与广西永福县长寿现象有关联。ApoEε3等位基因与永福长寿有关联,ApoEε3/ε3纯合子是最主要的永福长寿关联基因。
Objective To study the relationship between polymorphism of ApoE gene and longevity of Yongfu in long-lived elderly in Yongfu County of Guangxi. Methods A total of 319 longevity elders aged from 90 to 113 in Yongfu County of Guangxi and 168 longevity family members who were corresponding age controls were enrolled in this study by PCR-RFLP. 104 non-longevity natural population as the control group, ApoE genotyping, and to compare the longevity of the elderly and each group of genotypes, gene frequency distribution characteristics. Results (1) The frequencies of ApoE genotypes in longevity elderly in Yongfu County of Guangxi Province were highest with ε3 / 3 followed by ε2 / 3 and ε3 / 4, while those with ε2 / 4 and ε4 / 4 were the lowest. There was no significant difference between ApoE genotypes and gene frequencies in the longevity elderly group and their relatives age. (2) The genotype of ε3 / ε3 (82.4%; 75.0%) and the ε3 allele (90.44%, 85.17%) were significantly and positively correlated with those of the local population in the longevity group (P <0.05, P <0.01) . Conclusion ApoE gene polymorphism is associated with longevity in Yongfu County, Guangxi Province. The ApoEε3 allele is associated with Yongfu longevity, and the ApoEε3 / ε3 homozygote is the most important Yongfu longevity associated gene.
