目的:探讨胃癌组织HER2蛋白表达和基因拷贝数增加频率,以及蛋白表达与基因拷贝数之间的相关性。方法:分别采用免疫组织化学(IHC)和显色原位杂交(CISH)方法,检测80例青岛人胃癌组织中HER2蛋白表达和基因扩增情况。结果:HER2免疫组织化学0者51例、1+者12例、2+者12例、3+者5例。通过CISH分析:这组患者中HER2基因拷贝数扩增者共7例(8.8%),其中包括基因临界扩增3例(3.8%)。HER2的蛋白表达与基因拷贝数增加相关(P<0.05),这两个指标与肿瘤分化程度相关。HER2免疫组织化学3+和基因扩增结果一致,与乳腺癌相似。结论:这组患者中,具有较高的HER2蛋白表达和基因拷贝数增加的比率,基因扩增可能是其蛋白过表达的主要分子机制。
Objective: To investigate the frequency of HER2 protein expression and gene copy number increase in gastric cancer and the correlation between protein expression and gene copy number. Methods: Immunohistochemistry (IHC) and chromosomal in situ hybridization (CISH) were used to detect HER2 protein expression and gene amplification in 80 cases of gastric cancer in Qingdao. Results: HER2 immunohistochemistry 0 in 51 cases, 1 + 12 cases, 2 + 12 cases, 3 + 5 cases. By CISH analysis: HER2 gene copy number in this group of patients were a total of 7 cases (8.8%), including the gene critical amplification in 3 cases (3.8%). The protein expression of HER2 was correlated with the increase of gene copy number (P <0.05), which correlated with the degree of tumor differentiation. HER2 immunohistochemistry 3+ is consistent with the gene amplification results, similar to breast cancer. Conclusion: In this group of patients, the higher ratio of HER2 protein expression and gene copy number increase, gene amplification may be the main molecular mechanism of its protein overexpression.
