目的:探讨冠通方对急性心肌梗死(AMI)大鼠心肌梗死面积及心肌组织中CRP mRNA表达的影响。方法:20只成功造模的急性心肌梗死SD大鼠随机分为模型组、冠通方组,每组10只。空白组10只SD大鼠不作处理,假手术组10只开胸但不结扎冠脉。冠通方组冠通方灌胃,各组均正常饲料及饮水喂养,8周后处死大鼠。采用2,3,5-氯化三苯基四氮唑(TTC)法测量心肌梗死面积,逆转录聚合酶链反应(RT-qPCR)方法测定各组心肌组织中C反应蛋白(CRP)的mRNA表达结果。结果:(1)假手术组出现心肌损伤并未出现明显心肌梗死,冠通方组同模型组比较心肌梗死面积稍有下降,但无统计学意义(P>0.05)。(2)与空白对照组比较,模型组CRP mRNA表达量显著升高(P<0.01)。(3)与空白对照组比较,假手术组CRP mRNA表达量升高,无显著性差异(P>0.05)。(4)冠通方组与模型组相比,CRP mRNA表达量显著降低(P<0.05)。结论:CRP参与了心肌梗死后的炎症反应,中药冠通方对大鼠心肌梗死面积无明显影响,对心梗后CRP mRNA表达有一定抑制作用。
Objective: To investigate the effects of Guantong Prescription on myocardial infarct size and myocardial CRP mRNA expression in acute myocardial infarction (AMI) rats. Methods: Twenty SD rats with acute myocardial infarction were randomly divided into model group and Guan Tong Fang group, with 10 rats in each group. Ten rats in the blank group were not treated, and 10 in the sham-operated group were thoracotomy but did not ligate the coronary artery. Guan Tong Fang Guan Tong Tong prescription gavage, each group were fed with normal diet and water, rats were killed after 8 weeks. The area of myocardial infarction was measured by TTC method and the mRNA of C-reactive protein (CRP) in each group was determined by reverse transcription-polymerase chain reaction (RT-qPCR) Express the result. Results: (1) Myocardial infarction was not found in the sham operation group. The myocardial infarct size in the Guan Tong Fang group was slightly decreased compared with the model group, but there was no statistical significance (P> 0.05). (2) Compared with the blank control group, the expression of CRP mRNA in the model group was significantly increased (P <0.01). (3) Compared with the blank control group, the expression of CRP mRNA increased in the sham-operation group, but no significant difference (P> 0.05). (4) Compared with model group, the expression of CRP mRNA in Guan Tong Fang group was significantly lower (P <0.05). Conclusion: CRP is involved in the inflammatory reaction after myocardial infarction. Guanotong Decoction has no significant effect on myocardial infarct size in rats and CRP mRNA expression after myocardial infarction.
