分别采用种子切段、无菌苗真叶作外植体首次成功建立了大花飞燕草的组织培养和再生体系.结果表明:大花飞燕草种子切段和无菌苗真叶大多数是通过愈伤组织途径再生,也有极少数不经过愈伤组织阶段而直接再生出小植株。在合适的培养基上,种子切段和叶片两种外植体离体培养均能高效再生,平均每个外植体能分化出10—20个不定芽.种子切段培养的最适通用培养基为改良MS附加ZT3 mgL-1和 NAA 0.3 mg L-1;叶培养的最适通用培养基为 MS附加6-BA 3 mg L-1和 NAA 0.3 mg L-1.
The tissue culture and regeneration system of Pedicularis graminearum was successfully established for the first time using seed cuttings and sterile seedling leaves as explants. The results showed that most of the seedlings of A. lanceolata seeds and sterile seedlings were regenerated by callus, and a few of them regenerated directly without callus. On the appropriate medium, seed explants and leaves of both explants can be efficiently regenerated in vitro, with an average of 10-20 adventitious buds per explant. The optimum medium for seed-cut cultivation was ZT3 mgL-1 and NAA 0.3 mg L-1, respectively. The best medium for leaf culture was MS supplemented with 6-BA 3 mg L-1 and NAA 0 .3 mg L-1.
