目的:通过检测IL-27在小鼠实验性肠炎模型中的表达,探讨其在肠炎发病机制中的作用。方法:建立三硝基苯磺酸(TNBS)诱导的急性小鼠实验性肠炎模型,采用实时荧光定量PCR(qRT-PCR)及Western blot分别检测IL-27基因mRNA和蛋白在炎症肠黏膜中的表达。结果:TNBS成功诱导急性小鼠实验性肠炎,与对照组相比,TNBS处理的小鼠炎症评分明显增高,炎症肠黏膜中IL-27mRNA和蛋白表达亦明显增高,差异具有统计学意义。结论:IL-27在TNBS诱导的小鼠炎症肠黏膜中表达明显增高,提示IL-27可能参与小鼠肠道炎症的形成过程。
OBJECTIVE: To investigate the role of IL-27 in the pathogenesis of enteritis by detecting the expression of IL-27 in experimental mouse model of enteritis. Methods: Acute mouse experimental colitis induced by trinitrobenzene sulfonic acid (TNBS) was established. The expression of IL-27 mRNA and protein in inflammatory bowel mucosa were detected by real-time quantitative PCR (qRT-PCR) and Western blot expression. Results: TNBS induced acute experimental colitis in rats. Compared with the control group, TNBS-treated mice had significantly higher inflammatory score and IL-27 mRNA and protein expression in inflammatory intestinal mucosa. The difference was statistically significant. Conclusion: The expression of IL-27 in TNBS-induced mouse inflammatory bowel mucosa significantly increased, suggesting that IL-27 may be involved in the formation of intestinal inflammation in mice.
