目的 Meta分析TNF-α-308A/G单核苷酸多态性与皮肌炎相关性。方法在PubMed、EBSCO、EMBASE、中国学术期刊全文数据库(CNKI)、中国生物医学文献数据库(CBM)、万方数据库中查询肿瘤坏死因子α-308(TNF-α-308)位点多态性与皮肌炎相关性的文献。用Meta分析的方法分析基因型AA vs.GG,GA vs.GG,AA vs.GG+GA,GA+AA vs.GG和A vs.G在皮肌炎组与对照组中是否有差异。结果共纳入5篇文献进入本研究(462例皮肌炎患者,763例健康对照)。对总体人群进行Meta分析,发现TNF-α-308A等位基因与皮肌炎相关(OR=2.48,95%CI=2.00~3.07,P<0.001);将人群按照种族分层,发现-308A与高加索人群皮肌炎相关(OR=2.50,95%CI=1.99~3.14,P<0.001)。基因型AA vs.GG,GA vs.GG,AA vs.GG+GA,GA+AA vs.GG在皮肌炎组与对照组中均有统计学差异。结论 TNF-α-308A等位基因或基因型与高加索人群皮肌炎相关,TNF-α-308A等位基因或基因型可能是高加索人群皮肌炎的易感因素
Objective Meta analysis of TNF-α-308A / G single nucleotide polymorphism and dermatomyositis correlation. Methods Polymorphisms of tumor necrosis factor α-308 (TNF-α-308) loci were determined with PubMed, EBSCO, EMBASE, CNKI, CBM and Wanfang database. Dermatomyositis related literature. Meta-analysis was used to analyze whether genotypes AA vs.GG, GA vs.GG, AA vs.GG + GA, GA + AA vs.GG and A vs.G were different between the dermatomyositis group and the control group. Results A total of 5 articles were included in the study (462 dermatomyositis patients and 763 healthy controls). Meta-analysis of the overall population showed that the TNF-α-308A allele was associated with dermatomyositis (OR = 2.48, 95% CI = 2.00-3.07, P <0.001); stratifying the population by ethnicity found that -308A and Caucasians were associated with dermatomyositis (OR = 2.50, 95% CI = 1.99-3.14, P <0.001). The genotypes AA vs.GG, GA vs.GG, AA vs.GG + GA, GA + AA vs.GG were statistically different between the dermatomyositis group and the control group. Conclusions TNF-α-308A alleles or genotypes are associated with Caucasians. The TNF-α-308A allele or genotype may be a predisposing factor to Caucasian dermatomyositis
