液质联用串联质谱法测定人血浆中达托霉素浓度

目的建立一种迅速、简便和准确的液质联用串联质谱方法,用于测定人血浆中达托霉素(DPT)浓度。方法采用Zorbax SB-C_(18)column(100 mm×3.0 mm,3.5μm)色谱柱,以甲醇(含0.1%甲酸)-水(含0.1%甲酸)(70:30)为流动相,流速为0.5 mL·min~(-1),柱温为25℃;采用电喷雾离子源(ESI)正离子模式,多反应离子监测(MRM),[M+2H]~(2+)m/z 811.2→341.2定量DPT、[M+H]~+m/z 609.2→195.2定量内标利血平。结果 DPT和内标利血平的保留时间分别为2.99和1.27 min,DPT的线性范围为0.276~46.000 mg·L~(-1);提取回收率>70%;日内、日间RSD<5%(n=5)。结论本方法简便快速,适用于DPT制剂的新药临床研究和临床治疗患者血药浓度的常规监测。 Objective To establish a rapid, simple and accurate LC-MS / MS method for the determination of daptomycin (DPT) in human plasma. Methods The mobile phase consisted of methanol (containing 0.1% formic acid) and water (containing 0.1% formic acid) (70:30) as mobile phase on a Zorbax SB-C 18 column (100 mm × 3.0 mm, 0.5 mL · min -1 and the column temperature was 25 ℃. The electrospray ionization (ESI) positive ion mode, MRM, [M + 2H] 2+ m / z 811.2 → 341.2 Quantitative DPT, [M + H] ~ + m / z 609.2 → 195.2 Quantitative internal standard reserpine. Results The retention times of DPT and internal standard reserpine were 2.99 and 1.27 min respectively. The linear range of DPT was 0.276 ~ 46.000 mg · L -1. The recovery rate was> 70%. The intra-day and inter-day RSD was less than 5% (n = 5). Conclusion The method is simple and rapid, and is suitable for routine monitoring of plasma concentration of new drug clinical research and clinical treatment of DPT preparations.
