百慕大是一种难以培养的草坪草。本研究首次以百慕大成熟胚为外植体,通过胚性愈伤组织的发生进行植株再生。结果表明,百慕大成熟种子的出愈能力很强,且不同的基本培养基诱导效果相近,但初生愈伤组织呈无定型棉絮状,不能再生成株。采用提高培养基中蔗糖和琼脂浓度以及在培养基中附加适宜浓度ABA、CH和一定比例的生长素与细胞分裂素的方法,对初生愈伤组织实施继代改造,高频诱导出致密颗粒状的、具有高度再生能力的胚性愈伤组织,此类胚性愈伤组织在含有2.0mg/L6 BA、1.0mg/LKT和0.5mg/LNAA的分化培养基上再生频率较高。
Bermuda is a hard lawn grass. In this study, we first used Bermuda mature embryos as explants to regenerate embryogenic callus. The results showed that the ripening ability of Bermuda mature seeds was very strong, and the induction effect of different basic medium was similar. However, the primary callus showed amorphous cotton-like shape and could not be regenerated. The methods of raising the concentration of sucrose and agar in the medium, adding appropriate concentrations of ABA, CH and a certain proportion of auxin and cytokinin in the culture medium were used to transform the primary callus and the high frequency to induce the dense granular Of embryogenic callus with a high degree of regeneration. Such embryogenic callus had a higher frequency of regeneration on differentiation media containing 2.0 mg / L 6 BA, 1.0 mg / L KT and 0.5 mg / L NAA.
