为研究苯丙烷代谢关键酶——苯丙氨酸解氨酶(PAL)活性及相关基因表达在不同加勒比松变种/种源间的差异,本试验以盆栽加勒比松及湿地松苗木为材料,分析了其针叶PAL活性在不同季节及种源间的差异,结果表明:种源效应、环境温度及二者的互作效应均对PAL活性有显著作用,常温下,加勒比松14个种源PAL活性存在显著差异,但与对照湿地松差异较小;自然低温时,湿地松PAL活性显著增加,明显高于加勒比松除16与11外的其它种源,暗示PAL活性可能与加勒比松耐寒性存在一定的关系.进而利用定量PCR分析两个PAL同源基因在其中7个种源中的表达情况,发现常温下Pc PAL2与Pc PAL3的基因表达在种源间差异显著,且Pc PAL2表达与PAL活性高度正相关.本研究为利用PAL活性相关基因筛选加勒比松耐寒性种源,进一步遗传改良耐寒相关性状提供了理论依据.“,”In order to study the differences of phenylalanine ammonia-lyase (PAL) activity, a key enzyme of phenylaprapanoid metabolism, and related gene expression among the different Pinus caribaea variations/provenances, potted Pinus caribaea and P.elliottii were used as materials to analyze the differences of their needle PAL activities in different seasons and provenances.The results showed that the ambient temperature, provenance effect and the interaction effect between the two factors significantly affected PAL activities.At the normal temperature, the PAL activities were similar between P.caribaea and P.elliottii and significant variations were found among 14 provenances within P.caribaea.At the natural low temperature, the PAL activities of P.elliottii were significantly increased, which were obviously higher than provenances of P.caribaea except 16 and 11.The regulation patterns of the PAL activities implied that it might exist a certain relationship between cold hardiness of P.caribaea and the PAL activity.q PCR analysis was further applied to analyze the gene expression of two homologous genes Pc PAL2 and Pc PAL3among 7 provenances from P.caribaea and P.elliottii.The result showed that at the normal temperature gene expressions of Pc PAL2 and Pc PAL3 were significantly different in provenances, and Pc PAL2 gene expression had a strong positive correlation with PAL activity.This study might provide theoretical evidence for the screening of P.caribaea cold hardiness provenances by using PAL activity related genes and further genetic improvement of cold tolerance.
