目的:探讨筋骨草提取物(HBG)对乳腺癌及转移的抑制作用,及其对与乳腺癌侵袭和转移密切相关的基质金属蛋白酶及其抑制物的影响。方法:采用小鼠乳腺癌细胞株4T1原位接种制作乳腺癌自发转移模型,给予50~200 mg.kg-1HBG后评价各组肿瘤生长和肺转移情况;并用免疫印迹法测定肿瘤原位灶和转移灶MMP-2/9,TIMP-1/2的表达水平。结果:50~200 mg.kg-1 HBG呈剂量依赖性抑制乳腺癌生长和肺转移;下调MMP-2和MMP-9的表达;上调TIMP-1和TIMP-2表达。结论:筋骨草提取物通过调节乳腺癌细胞MMPs和TIMPs的表达而发挥抗乳腺癌及其转移的作用。
OBJECTIVE: To investigate the inhibitory effect of Radix Glabrae Extract (HBG) on breast cancer and its metastasis and its effect on matrix metalloproteinases and its inhibitors which are closely related to the invasion and metastasis of breast cancer. METHODS: Spontaneous metastasis model of breast cancer was established by in situ inoculation of mouse mammary carcinoma cell line 4T1, and the tumor growth and lung metastasis were evaluated after 50 ~ 200 mg.kg-1HBG administration. The tumor localization and lung metastasis were detected by immunoblotting Metastasis MMP-2/9, TIMP-1/2 expression levels. RESULTS: 50 ~ 200 mg.kg-1 HBG could inhibit the growth of breast cancer and lung metastasis in a dose-dependent manner; down-regulate the expression of MMP-2 and MMP-9; up-regulate the expression of TIMP-1 and TIMP- Conclusion: Radix extract can exert anti-breast cancer and its metastasis by regulating the expression of MMPs and TIMPs in breast cancer cells.
