本文研究了线粒体中甲状腺激素受体的亚微结构定位及其生物学和化学特性。从甲状腺正常的大鼠肝脏制备线粒体,再用毛地黄皂苷-Lubrol WX法和用水低渗溶解法分离线粒体的亚微成分,并进行对~(125)I-三碘甲状腺氨酸([~(125)T_3]的结合试验。结果发现,只有线粒体内膜上存在高亲和力、低结合量的物异结合部位,而线粒体外膜、膜内蛋白和基质蛋白都对~(125)-T_3无特异结合。在大鼠的肾、心肌、骨骼肌、小肠、脂肪组织及兔肾、人的肝和肾的线粒体、也获得相似结果。在幼年鼠(年令12天以前)的脑组织的线粒
In this paper, we studied the submicroscopic localization of thyroid hormone receptors in mitochondria and their biological and chemical properties. Mitochondria were prepared from the normal rat livers of the thyroid gland, mitochondrial submicroelements were isolated using the Digitonin-Lrolrol WX method and hypotonic solution with water, and the mitochondrial submicroelements were separated and assayed for ~ (125) I-triiodothyronine 125) T_3] .The results showed that only mitochondrial inner membrane with high affinity and low binding amount of different binding sites, and mitochondrial outer membrane, membrane proteins and matrix proteins were not specific to 125T3 Similar results were also obtained in the mitochondria of rat kidney, myocardium, skeletal muscle, small intestine, adipose tissue and rabbit kidney, human liver and kidney.When mitochondria were isolated from brain tissue of young rats (12 days old)
