ObjectiveTo observe the effect of short and long-term administration of tea polyphenols(TP) on +Gz induced peroxidative injury in mouse brain and TP's general toxicological action.
Methods①Short-term administration of TP: TP was given ip to male mice at dose of 100mg·kg-1·d-1, 3d ; for the controls 0.9% NaCl was given instead. After acceleration stress (+1 Gz,+3Gz,+9 Gz,+18 Gz for 15 min), malondialdehyde (MDA) formation in mouse brain was measured; ②Long-term administration of TP:TP was given orally to male mice at dose of 300 mg· kg-1·d-1,6 d·wk-1,4 wk;for the controls distilled water was given instead. After acceleration stress(+1 Gz,+18Gz for 15 min), MDA formation in mouse brain was determined;③General toxicological effect of TP:after long-term oral administration of TP, the body weight, organ indices, serum lipids, liver and renal functions and histomorphology of important organs were observed.
ResultsMDA formation in mouse brain was gradually and significantly increased with the elevation of exposed+Gz levels, while the lipid peroxidation was remarkably inhibited by short and long-term administration of TP. Toxicological examination showed that no harmful effect to mice after long-term administration of TP, and TP could significantly decrease the body weight and serum triglyceride level.
ConclusionNatural antioxidant TP had significantly protective effect on+Gz stress induced peroxidative injury in brain and was suitable for long-term administration.
