目的 探讨重组 380 0 0蛋白在结核病流行病调查和亚单位疫苗研制中的应用前景。方法 等电点测定、肽图分析和圆二色光谱分析研究重组 380 0 0蛋白的理化性质 ;豚鼠皮试 ,MTT法测外周血单核细胞增殖和巨噬细胞吞噬探讨重组 380 0 0蛋白在细胞免疫中所起的作用。结果 重组 380 0 0蛋白是酸性蛋白 ,其等电点为4 6 7;肽图分析表明其碱性氨基酸的数量与理论一致 ;重组 380 0 0蛋白的二级结构由α -螺旋 (32 6 % )、β 转角(31 6 % )和无规则卷曲 (35 8% )组成 ;它能诱发致敏豚鼠产生DTH反应 ;增强小鼠巨噬细胞吞噬功能 ;刺激30 8%健康者和 2 5 %所试结核病人的外周血单核细胞增殖。结论 重组 380 0 0蛋白可能在结核病流调中具有应用前景 ,并可作为亚单位疫苗候选者之一。
Objective To investigate the prospect of recombinant 380 0 0 protein in the investigation of tuberculosis epidemic and the development of subunit vaccine. Methods The isoelectric point, peptide mapping and circular dichroism spectroscopy were used to analyze the physico-chemical properties of the recombinant 380 0 protein. The guinea pig skin test and MTT assay were used to detect the proliferation of peripheral blood mononuclear cells and macrophage phagocytosis. The role of cellular immunity. Results The recombinant protein of 380 0 0 was an acidic protein with an isoelectric point of 4667. The peptide analysis showed that the number of its basic amino acids was consistent with the theory. The secondary structure of the recombinant protein was composed of α - helix (32 6% ), Β angle (31 6%) and irregular curl (35 8%). It induced DTH response in sensitized guinea pigs, increased phagocytosis of macrophages in mice, stimulated 30.8% of healthy subjects and 25% Test tuberculosis patients with peripheral blood mononuclear cell proliferation. Conclusion Recombinant 380 0 0 protein may have potential application in tuberculosis flow control and may serve as one of the subunit vaccine candidates.
