目的观察大鼠放射性肺损伤模型中血浆转化生长因子β1(TGF-β1)的变化以及灯盏细辛和曲尼斯特对其的影响。方法90只Wistar大鼠随机分为4组,空白组15只、单照组25只、曲尼斯特组25只、灯盏细辛组25只。单照组和两药物组用直线加速器产生的6 MV X射线照射右全胸,单次20 Gy。曲尼斯特组于照射前2周、灯盏细辛组于照射后开始每天灌胃给药,给药剂量曲尼斯特溶液50 mg/kg、灯盏细辛注射液50 mg/kg,持续到照射后2个月。空白组与单照组均灌胃相等体积的生理盐水。照射后每月取大鼠静脉血浆标本行TGF-β1检测,观察其变化情况。结果单照组血浆TGF-β1含量在照射后1个月开始增加,高峰在照射后第2个月,以后逐渐回落至一高水平。两药物组血浆TGF-β1含量2个月时明显低于单照组,但停药后仍出现高峰,高峰延至照射后第3个月。两药物组与单照组的TGF-β1含量均高于空白组,与肺损伤的程度一致。结论灯盏细辛和曲尼斯特能明显降低照射后血浆TGF-β1的含量,延长给药时间是否能增加疗效值得研究。
Objective To investigate the changes of plasma transforming growth factor β1 (TGF-β1) and the effects of breviscapine and transtain on it in a rat model of radiation-induced lung injury. Methods Ninety Wistar rats were randomly divided into four groups: 15 in blank group, 25 in monotherapy group, 25 in transthost group and 25 in fleabane group. Single and two drug groups were exposed to 6 MV X-rays generated by a linear accelerator in a right total chest with a single 20 Gy. In the transtain group, 2 weeks before irradiation, the Erigeron Breviscapus group was orally administered with gavage once a day for 50 mg / kg of Tranzstan solution and 50 mg / kg of Erigeron Breviscapus injection until irradiation 2 months. The blank group and the single group were given equal volume of normal saline. After irradiation, venous plasma samples from rats were taken every day for detection of TGF-β1 and their changes were observed. Results The level of plasma TGF-β1 in monotherapy group began to increase at 1 month after irradiation, and peaked at 2 months after irradiation, then gradually dropped to a high level. The levels of plasma TGF-β1 in the two drug groups were significantly lower than those in the single control group at 2 months, but remained peaked after stopping the treatment, with the peak extending to the 3rd month after irradiation. The levels of TGF-β1 in the two drug groups and the single group were higher than those in the blank group, which were consistent with the degree of lung injury. Conclusion Erigeron breviscapus and Tranister can significantly reduce the plasma levels of TGF-β1 after irradiation, prolonging the administration time whether to increase the efficacy worth studying.
