目的观察“三热论”方对糖尿病血管病变小鼠的保护作用及其机理。方法高脂饲料复合链脲佐菌素(STZ)诱导糖尿病小鼠模型,分为正常对照组、模型组、二甲双胍组、“三热论”方水提组、“三热论”方醇提组,各给药组灌胃给药体积为100 mL/kg。给药14 d,末次灌胃后禁食6 h,眼眶后静脉丛取血。测定小鼠血清中血糖、肌酐、尿素氮、一氧化氮(NO)等含量。结果 “三热论”方醇提组可显著降低模型小鼠血糖值(P<0.05);与模型组比较,“三热论”方水提组和醇提组小鼠肌酐与尿素氮含量降低,有显著差异(P<0.05,P<0.0);“三热论”方醇提物能显著降低模型小鼠血清中NO和一氧化氮合酶(NOS)含量(P<0.05)。结论 “三热论”方水提物和醇提物对糖尿病小鼠血管病发症有一定的防治作用。
Objective To observe the protective effect and mechanism of “Three heat Syndromes” on diabetic vascular lesions in mice. Methods Diabetic mice were induced by high-fat diet combined with streptozotocin (STZ) and divided into normal control group, model group, metformin group, Fang alcohol group, each administration group intragastric administration volume of 100 mL / kg. After 14 days, the rats were fasted for 6 hours after the last gavage, and blood was collected from the retro-orbital venous plexus. Determination of serum glucose, creatinine, urea nitrogen, nitric oxide (NO) and other content. Results Compared with the model group, creatinine and creatinine of mice in the three heat treatment groups and alcohol extraction group significantly decreased blood glucose level (P <0.05) (P <0.05, P <0.0). The alcoholic extract of “three heat” could significantly reduce the content of NO and NOS in the model mice (P <0.05, P < <0.05). Conclusion “Three heat theory ” square water extract and ethanol extract of diabetic mice have some prevention and treatment of vascular disease.
