目的研究柴胡、白芍及其配伍乙醇提取液对小鼠的抗抑郁作用和对四氯化碳(CCl4)所致小鼠肝损伤的保护作用。方法将60只雄性ICR小鼠随机分为空白组、模型组、阳性对照组、柴胡组、白芍组和柴胡-白芍组,每组10只。各组小鼠灌胃给药21 d后通过悬尾、强迫游泳等行为学实验研究各药的抗抑郁作用。另取6组雄性小鼠,分组同上,采用CCl4制作小鼠急性肝损伤模型,取上层血清检测天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)指标,并取肝脏组织做细胞形态学观察,以研究各药的保肝作用。结果各给药组均能显著缩短小鼠强迫游泳不动时间,拮抗利血平所致的运动能力下降、肛温变化、眼睑下垂,差异有统计意义(P<0.05或P<0.01),以柴胡-白芍组最佳;各给药组均能显著降低肝损伤小鼠血清AST,ALT含量,差异有高度统计意义(P<0.01),且可明显改善病理组织状况。结论柴胡、白芍乙醇提取液对小鼠具有明显的抗抑郁作用和保肝作用,且以柴胡-白芍配伍疗效最佳。
Objective To study the antidepressant effect of Bupleurum root, Radix Paeoniae Alba and its compatibility ethanol extract on mice and the protective effect against hepatic injury induced by carbon tetrachloride (CCl4) in mice. Methods Sixty male ICR mice were randomly divided into blank group, model group, positive control group, Bupleurum group, Radix Paeoniae Alba and Radix Bupleurum - Radix Paeoniae Alba - group with 10 in each group. The mice in each group were subjected to behavioral experiments such as tail-suspending and forced swimming for 21 days after gavage. The anti-depressive effects of each drug were studied. Another 6 groups of male mice were divided into groups as above. CCl4 was used to make a mouse model of acute liver injury, and the serum was collected to detect aspartate aminotransferase (AST) and alanine aminotransferase (ALT) Tissue morphological observation of cells to study the role of various drugs to protect the liver. Results Each administration group could significantly reduce forced immobilization time of mice, antagonize the decline of exercise capacity caused by reserpine, rectal temperature changes, ptosis, the difference was statistically significant (P <0.05 or P <0.01) Bupleuri - Radix Paeoniae Alba group was the best. Each treatment group could significantly reduce the serum AST and ALT levels in liver injury mice, the difference was highly statistically significant (P <0.01), and can significantly improve the pathological conditions. Conclusion Bupleurum root, ethanol extract of white peony root has obvious antidepressant effect and hepatoprotective effect in mice, and Bupleurum - Paeoniae radix compatibility best.
