体外培养小鼠腹腔巨噬细胞,分别加高脂血清及载药血清,测定不同培养液中NO、NOS、OX-LDL含量。并制备巨噬细胞,提取RNA,采用RT-PCR观察M-CSF mRNA表达。结果表明,调肝导浊中药可显著增强NOS活性,增加NO含量,抑制OX-LDL生成,并对M-CSF具有调节作用。提示调肝导浊中药具有良好的抗氧化修饰反应作用,从而有效防治AS形成发展。
The mouse peritoneal macrophages were cultured in vitro and hyperlipidemia serum and drug-loaded serum were added respectively to determine the content of NO, NOS, and OX-LDL in different culture fluids. Macrophages were prepared and RNA was extracted. M-CSF mRNA expression was observed by RT-PCR. The results showed that Tiaoganzhuzhu herbs can significantly enhance NOS activity, increase NO content, inhibit OX-LDL production, and have a regulatory effect on M-CSF. It is suggested that the liver-regulating and turbidity-removing Chinese medicine has a good anti-oxidation modification reaction, thus effectively preventing and controlling the formation and development of AS.
