目的 :研究p16及增殖细胞核抗原 (PCNA)在皮肤癌发病中的作用。方法 :应用免疫组化S P法检测抑癌基因p16蛋白及PCNA在 5 3例皮肤癌组织中的表达 ,并与癌旁组织和正常皮肤组织对比 ,结合皮肤癌的临床病理特征进行分析。结果 :皮肤癌组织中p16蛋白阳性表达率明显低于癌旁组织和正常皮肤组织 (P <0 0 5、P <0 0 1) ;p16蛋白的阳性表达率随着肿瘤浸润深度的加深而下降 (P <0 0 5 ) ;PTNMⅠ、Ⅱ期皮肤癌组织中p16蛋白阳性表达率明显高于PTNMⅢ、Ⅳ期 (P <0 0 5 )。PCNA指数随着肿瘤浸润深度的加深而增高 (P <0 0 5 ) ;PTNMⅢ、Ⅳ期皮肤癌组织中PCNA指数明显高于PT NMⅠ、Ⅱ期 (P <0 0 5 )。p16阴性组PCNA指数明显高于p16阳性组 (P <0 0 5 )。结论 :p16及PCNA表达与皮肤癌临床病理指标有关 ,皮肤癌的发生与演进是多因素作用的结果 ,两者的检测对估计皮肤癌的恶性度、判断预后及指导治疗有重要意义
Objective: To study the role of p16 and proliferating cell nuclear antigen (PCNA) in the pathogenesis of skin cancer. METHODS: Immunohistochemical SP method was used to detect the expression of tumor suppressor gene p16 protein and PCNA in 53 cases of skin cancer tissues, and compared with adjacent tissues and normal skin tissues, combined with clinical pathological features of skin cancer. Results: The positive expression rate of p16 protein in skin cancer tissues was significantly lower than that in adjacent tissues and normal skin tissues (P < 0.05, P <0.01). The positive expression rate of p16 protein decreased with the depth of tumor invasion. (P <0 05); The positive rate of p16 protein in PTNMI and stage II skin cancer tissues was significantly higher than that of PTN III and IV stages (P <0 05). The PCNA index increased with the depth of tumor invasion (P < 0.05). The PCNA index in PTNM III and IV skin cancer tissues was significantly higher than that of PT NMI and II (P <0 05). The PCNA index of the p16-negative group was significantly higher than that of the p16-positive group (P < 0 05). Conclusion : The expression of p16 and PCNA is related to the clinicopathological parameters of skin cancer. The occurrence and progression of skin cancer are the result of multiple factors. The detection of both is important for estimating the degree of malignancy of skin cancer, judging the prognosis and guiding the treatment.
