目的:建立血浆中维生素A的高效液相色谱快速测定方法。方法:在避光条件下,血浆加入内标后振荡混匀,经正己烷萃取后离心,取上清液氮气吹干,加入无水乙醇溶解,HPLC测定。结果:在0.034μg/ml~0.68μg/ml的浓度范围内线性关系良好,r=0.9998,方法回收率88.2%~111.8%,最低检出限0.6 ng。结论:该方法操作简便、快速、准确,能够满足日常大批量快速分析要求。
Objective: To establish a rapid determination method of vitamin A in plasma by high performance liquid chromatography. Methods: Under the dark conditions, the plasma was added with internal standard and then mixed by shaking. After extracting with n-hexane, centrifugation was carried out. The supernatant was dried under nitrogen, dissolved in absolute ethanol and determined by HPLC. Results: The linearity was good in the concentration range of 0.034μg / ml ~ 0.68μg / ml, r = 0.9998. The recoveries were 88.2% ~ 111.8% with the lowest detection limit of 0.6 ng. Conclusion: The method is simple, rapid and accurate, and can meet the requirements of daily mass rapid analysis.
