目的:研究白血病抑制因子(Leukemia inhibitory factor,LIF)在子宫内膜异位症(简称内异症,Emdometrio-sis,EMS)患者种植窗期子宫内膜排卵期卵泡液中的表达情况,探讨其与EMS患者IVF-ET(In vitro fertilization-embryotrans-fer)妊娠率下降的关系。方法:利用23例不孕合并EMS患者签订知情同意书的情况下IVF前一周期种植窗期取子宫内膜,取卵日收集废弃的卵泡液。采用免疫组化SABC法检测内膜LIF,双抗体酶联免疫吸附法(ELISA)测定取卵日卵泡液中LIF含量,对照组是47例非EMS患者。分析LIF的表达情况以及与胚胎发育、妊娠的关系。结果:非EMS患者与EMS患者获卵数(13.86±8.25)和(9.43±7.12)、受精率(76.05%和60.59%)、可用胚率(54.28%和34.47%)、胚胎种植率26.53%(26/98)和8.51%(4/27)、临床妊娠率51.06%(24/47)和17.39%(4/23),差异均有统计学意义(P<0.05);LIF在取卵期卵泡液中分别为(25.10±10.16)pmol/L和(14.68±5.54)pmol/L,差异有统计学意义(P<0.05)。LIF在种植窗期子宫内膜表达情况,EMS组显著低于非EMS组(P<0.05)。结论:EMS患者种植窗期的子宫内膜、取卵日卵泡液中LIF表达均下降,可能与EMS导致不孕有相关性。
Objective: To investigate the expression of leukemia inhibitory factor (LIF) in follicular fluid of endometrial ovulation during the implantation of endometrium in patients with endometriosis (EMD) And EMS patients IVF-ET (In vitro fertilization-embryotrans-fer) pregnancy rate decreased relationship. Methods: Endometrial endomembrane was collected from the implantation window of the previous period of IVF in 23 cases of infertility with EMS, and discarded follicular fluid was collected on ovulation day. Immunofluorescence SABC method was used to detect endometrial LIF. ELISA was used to determine LIF content in follicular fluid of ovulation day. The control group was 47 non-EMS patients. Results: The number of oocytes (13.86 ± 8.25) and (9.43 ± 7.12), fertilization rates (76.05% and 60.59%), embryo rates (54.28% and 34.47%) and embryo implantation rates of 26.53% The clinical pregnancy rates were 51.06% (24/47) and 17.39% (4/23), respectively, with statistical significance (P <0.05) (25.10 ± 10.16) pmol / L and (14.68 ± 5.54) pmol / L respectively, the difference was statistically significant (P <0.05). The expression of LIF in the implantation window was significantly lower in the EMS group than in the non-EMS group (P <0.05). CONCLUSION: The expression of LIF in the endometrium and ovulation follicular fluid of EMS patients during the implantation window is decreased, which may be related to the EMS infertility.
