目的观察不同浓度去甲肾上腺素 ( NA,10 - 1 0~ 10 - 6 m ol/ L)对大鼠体外抗体生成的影响 ,并初步探讨其作用机制。方法在体外用绵羊红细胞 ( SRBC)刺激大鼠肠系膜淋巴结 B细胞转化成抗体形成细胞 ( AFC) ,然后检测其抗体生成量。结果 110 - 1 0、10 - 9、10 - 8和 10 - 7mol/ L NA都能显著抑制体外抗体生成 ,其中 10 - 8mol/ L NA的作用最强。2 β受体激动剂异丙基肾上腺素亦能明显减弱体外抗体生成。3 α受体拮抗剂酚妥拉明和 β受体拮抗剂心得安不能单独阻断 NA抑制体外抗体生成的作用 ,而两者共同作用时 ,能完全阻断 NA的作用。4NA仍能抑制先用 SRBC刺激 2 4h或 48h后的淋巴细胞的抗体生成 ,但不能抑制用 SRBC刺激 72 h或 96 h后的淋巴细胞的抗体生成。结论 NA可非浓度依赖性地抑制大鼠的体外抗体生成 ,此作用可能由淋巴细胞上的 α和 β受体共同介导 ,且 NA可能主要影响淋巴细胞转化的中期阶段
Objective To observe the effect of different concentrations of norepinephrine (NA, 10 - 10 ~ 10 -6 mol / L) on the antibody production in vitro in rats and its mechanism. Methods In vitro, B cells of mesenteric lymph nodes of rats were stimulated with SRBC to form antibody-producing cells (AFCs), and then their antibody production was measured. Results Both 110 - 1, 10 - 9, 10 - 8 and 10 - 7 mol / L NA could significantly inhibit the antibody production in vitro, of which 10 - 8 mol / L NA had the strongest effect. 2 β-agonist isoproterenol also significantly attenuated in vitro antibody production. 3 α receptor antagonist Phentolamine and β receptor antagonist propranolol can not block NA alone inhibit the antibody production in vitro, and the two together, can completely block the role of NA. 4NA still inhibited the antibody production by lymphocytes stimulated with SRBC for 24 hours or 48 hours but not the antibody production by lymphocytes stimulated with SRBC for 72 hours or 96 hours. Conclusion NA can inhibit the antibody production in vitro in a concentration-dependent manner in rats. This effect may be mediated by α and β receptors on lymphocytes, and NA may mainly affect the metaphase of lymphocyte transformation
