目的:优选XDA-7型大孔吸附树脂纯化白子菜总多糖的条件,为该有效部分的充分利用提供参考。方法:采用Plackett-Burman试验筛选影响总多糖纯化工艺的显著性因素,以总多糖保留率及蛋白质去除率为因变量,上样量、流速、洗脱体积、径高比、上样液质量浓度及p H为自变量,通过响应面试验优选白子菜总多糖的纯化条件。结果:上样量、流速和洗脱体积为影响总多糖纯化效果的显著性因素,在上样量0.93 g·g-1(生药材-树脂),流速1.36 m L·min-1,洗脱剂用量4.3 BV的条件下,白子菜总多糖保留率93.08%,蛋白质去除率49.13%。二者与预测值的相对误差分别为3.49%,2.09%。结论:XDA-7型大孔吸附树脂能较好地保留总多糖、去除蛋白质,适用于白子菜总多糖的纯化处理。
OBJECTIVE: To optimize the conditions for the purification of total polysaccharide of X. radix XDA-7 macroporous resin for reference to make full use of the effective fraction. Methods: The Plackett-Burman test was used to screen the significant factors influencing the purification process of total polysaccharides. The total polysaccharide retention rate and protein removal rate were used as dependent variables, loading volume, flow rate, elution volume, aspect ratio, And p H as independent variables, through the response surface test to optimize the purification conditions of the total polysaccharide of cabbage. Results: The sample volume, the flow rate and the elution volume were the significant factors affecting the purification of total polysaccharides. When the sample volume was 0.93 g · g-1 (crude drug-resin) and the flow rate was 1.36 m L · min-1, Under the condition of 4.3 BV, the total polysaccharide retention rate of Chaetomium japonicum was 93.08% and the protein removal rate was 49.13%. The relative errors between the two and predicted values were 3.49% and 2.09% respectively. Conclusion: The macroporous resin XDA-7 can retain the total polysaccharide better and remove the protein, which is suitable for the purification of total polysaccharide of Chardium.
