采用固体培养和液体培养方法研究了鸡菌的培养特性以及培养基、pH值、酵母膏对其菌丝生长的影响。结果表明:鸡菌在普通固体培养基中生长缓慢,菌落较小,在培养后期,菌株间存在3种明显的菌落类型即“菌丝型”、“孢子型”、“中间型”,产孢量和形态也有较大差异。在液体培养条件下鸡菌的生长较快,培养基、pH值、酵母膏浓度对液体菌丝生长存在很大影响。各供试菌株在液体摇床培养中的最佳培养条件基本一致:即最适pH值为4.5;酵母膏最佳质量浓度为0.2 g/dL;理想培养基为PSDYB。
The solid culture and liquid culture methods were used to study the culture characteristics of Chlamydia trachomatis as well as the effects of medium, pH and yeast extract on its mycelial growth. The results showed that there were three kinds of obvious colony types, ie “mycelial type”, “spore type”, “intermediate type” and “middle type” among the strains during the later stage of culture. Spore size and morphology are also quite different. Under the conditions of liquid culture, the growth of C: giomyces was rapid, and the medium, pH value and the concentration of yeast extract had great influence on the mycelium growth. The best culture conditions of the tested strains in the liquid shaker culture were basically the same: the optimum pH was 4.5; the optimum concentration of yeast extract was 0.2 g / dL; the ideal medium was PSDYB.
