以人类5~6月龄胎儿各种组织为原料,用琼脂糖凝胶电泳法分离各种组织LDH同工酶,用酶联扫描法测定LDH同工酶的相对活力。通过酶谱分析,在男女胎儿各种组织间发现相同组织有相同的LDH同工酶酶谱;不同组织有相同的LDH同工酶酶谱;相同的组织有不同的LDH同工酶酶谱以及不同组织有不同的LDH同工酶酶谱。LDH同工酶酶谱的分析,提供了一个从分子水平上揭示高等脊椎动物基因表达调控机理的研究途径。
Human fetal tissue from 5 to 6 months of age was used as raw material. The LDH isoenzymes of various tissues were separated by agarose gel electrophoresis. The relative activity of LDH isoenzymes was determined by enzyme-linked immunosorbent assay. By zymogram analysis, the same LDH isozyme zymogram was found in the same tissues between male and female fetuses; the same LDH isozyme zymogram was found in different tissues; the same tissue had different LDH isoenzyme zymograms and Different organizations have different LDH isozymes. The analysis of the LDH isoenzyme zymogram provides a molecular approach to reveal the regulatory mechanisms of higher vertebrate gene expression.
