目的 研究B7基因修饰的肝癌细胞疫苗刺激产生细胞因子 (IL 2 ,TNF α和G CSF)的能力。方法 应用逆转录病毒载体 ,将B7 1、B7 2基因导入小鼠肝癌细胞Hepal 6细胞株中 ,获高表达B7分子的阳性细胞克隆 ,经丝裂霉素C(MMC)处理 ,制备肿瘤细胞疫苗 (TCV) ,观察TCVB7体内外对细胞因子IL 2 ,TNF α和C CSF产生的影响。结果 (1 )TCVB7体外能刺激脾细胞分泌细胞因子IL 2 ,TNF α和G CSF ;(2 )TCVB7免疫的小鼠 ,其血清IL 2和G CSF水平升高。结论 B7基因修饰的肝癌细胞疫苗能刺激产生细胞因子IL 2 ,TNF α和G CSF。
Objective To investigate the ability of B7 gene-modified hepatoma cell vaccine to stimulate the production of cytokines (IL 2, TNF α and G CSF). Methods B7 1 and B7 2 genes were introduced into Hepal 6 cell line of mice by retroviral vector and positive clones with high expression of B7 molecule were obtained and treated with mitomycin C (MMC) to prepare tumor cell vaccine (TCV) to observe the effects of TCVB7 on the production of cytokines IL 2, TNF α and C CSF in vitro and in vivo. Results (1) TCVB7 could stimulate the secretion of cytokines IL 2, TNFα and G CSF in splenocytes in vitro; (2) The TCVB7 immunized mice had elevated serum IL 2 and G CSF levels. Conclusion B7 gene modified hepatocarcinoma cell vaccine can stimulate the production of cytokines IL 2, TNF α and G CSF.
