目的:拟建立一种能预防和治疗移植后病毒感染的抗原呈递细胞( antigen presenting cell,APC)。方法:利用人的脐带血单个核细胞( CB derived mononuclear cells, CBMC),用 EB病毒 (Epstein Barr virus, EBV)感染转化成 B淋巴母细胞细胞系( B lymphoblastoid cell lines, BLCL),再用编码有 CMV.pp65蛋白基因的逆转录病毒转染,最后经 Western blot和免疫组化检测,证实该株细胞同时表达 EBV和巨细胞病毒 (cytomegalovirus, CMV)特异性蛋白。结果:获得了能同时表达 EBV和 CMV特异性蛋白的细胞株。结论:上述方法可以建立能同时表达 EBV和 CMV特异性蛋白抗原的细胞系。
Objective: To establish an antigen presenting cell (APC) that can prevent and treat post-transplant virus infection. Methods Human CB-derived CB cells were transformed into B lymphoblastoid cell lines (BLCL) by infection with Epstein Barr virus (EBV) The retrovirus transfected with the CMV.pp65 protein gene was confirmed by Western blot and immunohistochemistry. The results showed that both EBV and cytomegalovirus (CMV) specific proteins were expressed in this strain. Results: Cell lines expressing both EBV and CMV-specific proteins were obtained. Conclusion: The above method can establish a cell line that can express both EBV and CMV specific protein antigens simultaneously.
