Objective To study the expression of NaV1.6 mRNA and protein in the hippocampus after traumatic brain injury (TBI). Methods Adult SD rats were subjected to hydrocephalus. Rats were sacrificed at 2, 12, 24 and 72 h after injury. Real-time PCR and Western blotting were performed on the injured side of the hippocampus to detect the mRNA and protein expression of NaV1.6. Results The expression of NaV1.6 mRNA was significantly up-regulated at 2 h after traumatic brain injury in rats (P <0.001), and reached the highest level at 12 h after injury. The expression of NaV1.6 protein was significantly up-regulated to 72 h after injury (P <0.01) ). Conclusions TBI can up-regulate the expression of NaV1.6 mRNA and protein, which may be one of the molecular bases for the dysfunction of sodium channel and the excitotoxicity induced by TBI.
