目的通过检测食管癌患者和正常健康体检者血清中白细胞介素12(Interleukin-12,IL-12)、IL-16和IL-17表达水平,探讨3种细胞因子在食管癌中的作用和临床意义。方法随机选取2013年6月—2014年6月在南充市中心医院就诊的食管癌患者51例作为试验组、40例正常健康体检者作为对照组,收集受试者外周静脉血血清,酶联免疫吸附实验(enzyme-linked immunosorbent assay,ELISA)检测受试者IL-12、IL-16和IL-17血清表达水平,利用统计学软件SPSS 16.0,采用独立样本t检验、单向方差分析(One-way ANOVA)和偏相关分析(Partial)对数据进行详细分析。结果 IL-12、IL-16和IL-17在食管癌患者血清中表达水平均高于正常健康体检者(P<0.01),食管癌患者血清IL-12表达水平明显高于IL-16和IL-17(P<0.01),健康体检者血清IL-12和IL-16表达水平明显高于IL-17(P<0.01);在去除IL-16影响的条件下,食管癌患者血清中IL-12和与IL-17表达水平呈显著正相关(P<0.01);在去除IL-12影响的条件下,食管癌患者血清中IL-16和IL-17表达水平呈显著正相关(P<0.01)。结论与正常健康对照组比较,IL-12、IL-16和IL-17在食管癌患者血清中均呈高表达水平,3种细胞因子在食管癌的发生发展过程中具有重要作用,推测在食管癌发生发展过程中存在这3种细胞因子免疫调控失衡,3种细胞因子间的相互影响可能存在复杂的信号传导通路。
Objective To investigate the role of three cytokines in the pathogenesis of esophageal cancer by detecting the expression levels of Interleukin-12 (IL-12), IL-16 and IL-17 in patients with esophageal cancer and normal healthy subjects significance. Methods Totally 51 patients with esophageal cancer who were treated in Nanchong Central Hospital from June 2013 to June 2014 were selected as the experimental group and 40 normal healthy subjects as the control group.The peripheral venous blood serum and enzyme-linked immunosorbent assay The levels of IL-12, IL-16 and IL-17 in serum were detected by enzyme-linked immunosorbent assay (ELISA). Using SPSS 16.0 statistical software, way ANOVA and partial correlation analysis. Results The expression levels of IL-12, IL-16 and IL-17 in serum of patients with esophageal cancer were significantly higher than those of healthy controls (P <0.01). The expression of IL-12 in esophageal cancer patients was significantly higher than that of IL-16 and IL- -17 (P <0.01). The serum levels of IL-12 and IL-16 in healthy subjects were significantly higher than those in IL-17 (P <0.01). Under the influence of IL-16, the levels of IL- (P <0.01). The level of IL-16 and IL-17 in serum of patients with esophageal cancer was positively correlated with the level of IL-17 (P <0.01) ). Conclusion Compared with normal healthy control group, IL-12, IL-16 and IL-17 are highly expressed in the serum of esophageal cancer patients. The three cytokines play an important role in the occurrence and development of esophageal cancer. There are imbalances in the immunological regulation of these three cytokines during the carcinogenesis and development. The interaction between the three cytokines may have complex signal transduction pathways.
