目的研究Th17/Treg细胞在肝癌组织及癌旁组织中的表达,探讨其在肝细胞癌肿瘤微环境中的变化及意义。方法选取2015年1月-2016年4月因肝细胞癌在本院普外科住院的患者28例,采用流式细胞仪检测肝细胞癌肿瘤组织(TIL)和癌旁组织(NIL)的Th17/Treg细胞频数,实时定量逆转录PCR(RT-PCR)检测IL-17和Foxp3的mRNA表达水平。结果 TIL组的Th17细胞和Foxp3+CD4+CD25+Treg细胞百分率明显高于NIL组,差异有统计学意义(P<0.01)。TIL组的IL-17和Foxp3的mRNA表达水平明显高于NIL组,差异有统计学意义(P<0.01)。经单因素相关分析Pearson检验,TIL组及NIL组中Th17细胞百分率与IL-17的mRNA表达水平呈正相关(r值分别为0.754、0.812,P<0.05);TIL组及NIL组中Foxp3+CD4+CD25+Treg细胞百分率与Foxp3的mRNA表达水平呈正相关(r值分别为0.832、0.784,P<0.05)。结论 Th17/Treg细胞可能参与了肝细胞癌肿瘤发展与浸润。
Objective To study the expression and significance of Th17 / Treg cells in hepatocellular carcinoma tissues and adjacent tissues. Methods Twenty-eight patients hospitalized with hepatocellular carcinoma in our department from January 2015 to April 2016 were enrolled in this study. The Th17 / T lymphocyte subsets of hepatocellular carcinoma (TIL) and adjacent non-cancerous tissue (NIL) were detected by flow cytometry Treg cell frequency, real-time quantitative reverse transcription PCR (RT-PCR) detection of IL-17 and Foxp3 mRNA expression levels. Results The percentage of Th17 cells and Foxp3 + CD4 + CD25 + Treg cells in TIL group was significantly higher than that in NIL group (P <0.01). The mRNA expression levels of IL-17 and Foxp3 in TIL group were significantly higher than those in NIL group (P <0.01). The Pearson test showed that there was a positive correlation between the percentage of Th17 cells and the mRNA expression of IL-17 in TIL group and NIL group (r = 0.754,0.812, P <0.05, respectively). In TIL group and NIL group, Foxp3 + CD4 + CD25 + Treg cells was positively correlated with Foxp3 mRNA expression (r = 0.832,0.784, P <0.05). Conclusion Th17 / Treg cells may be involved in the development and infiltration of hepatocellular carcinoma.
