目的 :观察极低密度脂蛋白 (VL DL)与低密度脂蛋白 (L DL)是否能刺激肝细胞纤溶酶原激活物抑制物 - 1(PAI- 1)的分泌。方法 :将不同浓度的 VL DL 与 L DL 加到培养的 Hep G2细胞 ,应用酶联免疫吸附法测定细胞上清液中的 PAI- 1抗原 ,采用逆转录 -多聚酶链式反应 (RT- PCR)观测 Hep G2细胞中 PAI- 1m RNA的表达。结果 :VL DL能刺激 Hep G2细胞 PAI- 1抗原的分泌并具剂量效应关系 ,同时 PAI- 1m RNA表达增加。相反 L DL则不能刺激细胞 PAI- 1基因的表达。结论 :VL DL能刺激肝细胞 PAI- 1基因表达 ,VL DL的这种作用可能参与了致动脉粥样硬化的病理过程。
Objective: To observe whether very low density lipoprotein (VL DL) and low density lipoprotein (L DL) can stimulate the secretion of plasminogen activator inhibitor - 1 (PAI - 1) in hepatocytes. Methods: Different concentrations of DL DL and L DL were added into cultured Hep G2 cells. The PAI-1 antigen in the cell supernatant was determined by enzyme-linked immunosorbent assay (ELISA), and the reverse transcriptase-polymerase chain reaction (RT-PCR) PAI-1m RNA expression in Hep G2 cells was observed. Results: VL DL stimulated the secretion of PAI-1 antigen in Hep G2 cells with a dose-response relationship and the expression of PAI-1 mRNA increased. Conversely, L DL does not stimulate the expression of the PAI-1 gene in cells. CONCLUSION: VL DL can stimulate the expression of PAI-1 gene in hepatocytes. This effect of VL-DL may be involved in the pathological process of atherosclerosis.
