目的:探讨贯叶连翘中金丝桃素的酶提取法的最优酶及最佳工艺。方法:在果胶酶、木聚糖酶、β-葡聚糖酶、复合酶SPE-007A中筛选出最优酶,并以高效液相色谱法测定金丝桃素含量为指标,继考察浸提倍数、加酶量、pH值、温度、酶解时间等因素对提取工艺的影响,并用正交试验优选提取条件,从而得到最佳工艺。结果:最优酶为果胶酶,优选出的工艺为pH4.6、酶用量1.5%、酶解温度50℃、酶解时间5 h、酶解液浸提倍数15倍。结论:该工艺高效、稳定,可用于今后对贯叶连翘的研究。
Objective: To investigate the optimal enzyme and the best technology of enzyme extraction of Hypericin from Hypericum perforatum. Methods: The best enzyme was screened out in pectinase, xylanase, β-glucanase and synthase SPE-007A. Hypericin content was determined by HPLC. Multiple times, enzyme dosage, pH value, temperature, enzymolysis time and other factors on the extraction process, and the optimum extraction conditions using orthogonal test to get the best process. Results: The optimal enzyme was pectinase. The optimized process was pH4.6, enzyme dosage 1.5%, enzymolysis temperature 50 ℃, enzymolysis time 5 h, digestion times 15 times. Conclusion: The process is efficient and stable and can be used for the future study of Hypericum perforatum.
