山金柑(Forrunella hindisti Swingle)种子在MT培养基上培养50天,可从幼苗子叶下0.1—2.0厘米处产生胚性愈伤组织,并能在无激素的基本培养基中继代保存,仍有胚性再生能力。取自田间植株的种子,其试管苗产生胚性愈伤组织的频率为16.7%,显著地低于温室盆栽树种子试管苗愈伤组织的发生频率37.1%,不同年份之间这种频率没有显著差异。产生愈伤组织的实生苗高度只有未产生愈伤组织苗高的66.4%,叶片数也只有后者的1/3,将子叶以下0.5厘米的茎段在基本培养基上进行离体培养,38.5%的外植体产生了胚性愈伤组织。
Forrunella hindisti Swingle seeds are cultured on MT medium for 50 days and produce embryogenic callus from 0.1-2.0 cm below seedling cotyledons and can be subcultured in hormone-free minimal medium with Embryogenic regeneration ability. The frequency of embryogenic callus from test-tube seedlings collected from field plants was significantly lower than that of greenhouse seedlings (37.1%), with no significant difference between years difference. The callus-producing seedling height was only 66.4% of the callus did not produce callus, the number of leaves is only 1/3 of the latter, the 0.5 cm cotyledon below stem segments were cultured in vitro on minimal medium, 38.5 % Of explants produced embryogenic callus.
