目的:探讨喉鳞状细胞癌(LSCC)中Skp2和p27蛋白的表达与患者临床病理和预后的关系以及两种蛋白之间的相关性。方法:用免疫组织化学SP法检测20例正常喉黏膜(NLT)、27例LSCC癌前病变不典型增生(LPL)和70例LSCC组织中Skp2和p27蛋白的表达情况。结果:Skp2蛋白在NLT、LPL和LSCC中的阳性表达率分别为10.00%(2/20),37.04%(10/27)和41.43%(29/70),后两者的阳性率显著高于前者(均P<0.05)。Skp2蛋白表达与临床分期及淋巴结转移显著正相关(P<0.05)。p27蛋白在NLT和LSCC中阳性率分别为85.00%(17/20),48.57%(34/70),两者比较,差异有统计学意义(P<0.01)。p27蛋白表达与颈淋巴结转移呈负相关(P<0.05)。Skp2蛋白在LSCC中表达与p27蛋白呈负相关(P<0.05)。Skp2表达阳性而p27表达阴性的LSCC患者生存率明显低于其他患者(P<0.05)。结论:Skp2可能通过对细胞周期抑制因子p27蛋白的降解而参与LSCC的发生、发展,联合检测Skp2及p27蛋白的表达对预测LSCC患者的预后具有重要意义。
Objective: To investigate the relationship between the expression of Skp2 and p27 proteins in clinic, pathology and prognosis of patients with laryngeal squamous cell carcinoma (LSCC) and the correlation between the two proteins. Methods: The expressions of Skp2 and p27 proteins in 20 cases of normal laryngeal mucosa (NLT), 27 cases of LSCC (atypical hyperplasia of LSCC) and 70 cases of LSCC were detected by immunohistochemical SP method. Results: The positive expression rates of Skp2 protein in NLT, LPL and LSCC were 10.00% (2/20), 37.04% (10/27) and 41.43% (29/70), respectively. The positive rates of Skp2 protein were significantly higher The former (all P <0.05). Skp2 protein expression was positively correlated with clinical stage and lymph node metastasis (P <0.05). The positive rates of p27 protein in NLT and LSCC were 85.00% (17/20) and 48.57% (34/70), respectively. There was significant difference between them (P <0.01). The expression of p27 protein was negatively correlated with cervical lymph node metastasis (P <0.05). The expression of Skp2 protein in LSCC was negatively correlated with p27 protein (P <0.05). The survival rate of patients with positive Skp2 expression but negative p27 expression was significantly lower than that of other patients (P <0.05). Conclusion: Skp2 may be involved in the occurrence and development of LSCC through the degradation of cell cycle inhibitor p27, and the combined detection of Skp2 and p27 protein expression is of great significance in predicting the prognosis of patients with LSCC.
